Quality control for lyophilized materials includes visual inspection, residual moisture measurement, and reconstitution testing. Cake appearance can reveal process problems such as collapse, shrinkage, or meltback, although appearance alone does not prove potency. Residual moisture is commonly measured by Karl Fischer titration or by loss on drying. Reconstitution time is checked because a slow or incomplete dissolve can indicate a change in pore structure. Stability studies track these attributes over time under defined temperature and humidity conditions.
Analytical methods for lyophilized solids must account for the low moisture content and the fragile cake. Karl Fischer titration is widely used for water content, while near-infrared spectroscopy can measure moisture non-destructively in sealed containers. X-ray diffraction and modulated differential scanning calorimetry help identify crystalline or amorphous phases. Residual solvent analysis may be needed if organic solvents were used during formulation. The combination of these methods supports batch release and long-term stability assessment.
The physics of lyophilization couples heat transfer, mass transfer, and phase behavior. Sublimation requires a vapor pressure difference between the ice front and the chamber, and the dried layer adds resistance to vapor flow. Amorphous formulations are characterized by a glass transition temperature of the maximally freeze-concentrated solute, often denoted Tg'. Crystalline bulking agents can provide structure, while amorphous excipients stabilize labile components. Open questions remain about spatial heterogeneity, edge effects, and how laboratory cycles scale to production.
Lyophilization is a drying process in which a solvent, usually water, is removed from a frozen material by sublimation under reduced pressure. The material is first solidified, then placed under vacuum so that ice transitions directly to vapor without a bulk liquid phase. This approach suits heat-sensitive substances that would degrade during conventional evaporation. Primary drying removes unbound ice, while secondary drying reduces water that remains adsorbed to the solid matrix. The result is a porous, lightweight solid that can be reconstituted later.
A typical cycle begins with freezing, sometimes including an annealing step to control ice crystal size. Freezing conditions influence the pore network that later allows vapor escape. During primary drying, shelf temperature and chamber pressure are set so heat enters the product while its temperature stays below the collapse or eutectic point. Secondary drying then raises the shelf temperature to desorb bound water and lower residual moisture. Cycle design depends on formulation, fill volume, container type, and equipment capability.
| Property | Value | Notes |
|---|---|---|
| Storage temperature | 2–8 °C or 20–25 °C | Depends on product stability; some require frozen storage. |
| Moisture content | 0.5–3% w/w | Higher values may reduce stability; target set per product. |
| Moisture method | Karl Fischer titration | Coulometric for low levels; volumetric for higher levels. |
| Cake appearance | Uniform, intact, no collapse | Visual inspection is qualitative and not a potency measure. |
| Reconstitution time | Seconds to several minutes | Depends on cake density, excipients, and diluent. |
Misconceptions about lyophilization include the idea that dried products are indefinitely stable or that vacuum sealing eliminates all degradation. Chemical reactions can continue in the solid state, and some proteins lose activity even at low moisture. Another misconception is that any freeze-dryer cycle can be scaled by time alone; heat and mass transfer differ with equipment and load. Open questions remain about predicting long-term stability from short accelerated studies, particularly for amorphous formulations. Real-time stability data remain the standard for shelf-life assignment.
After lyophilization, the dried product is often sealed under vacuum or an inert gas to limit moisture and oxygen exposure. Vials, stoppers, and seals must maintain their barrier throughout shelf life. Storage temperature depends on product sensitivity: some cakes tolerate controlled room temperature, while labile biologics require refrigeration. Humidity is a critical variable because dried cakes are hygroscopic and can absorb water when exposed to air. Handling procedures therefore limit open-vial time and use desiccated environments for sampling.
Equipment for lyophilization includes a vacuum chamber, temperature-controlled shelves, a condenser, and a vacuum pump. A refrigeration system cools the shelves and condenser below the product's freezing point. Process monitoring often uses Pirani and capacitance manometers, thermocouples, and resistance sensors. Cycle development balances product quality with time and energy use. Some products are annealed during freezing to improve crystallization of bulking agents. Open questions remain about scaling cycles between laboratory, pilot, and production freeze-dryers.
Lyophilization, also called freeze-drying, removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts free water into ice and may also produce a glassy phase. Primary drying then lowers chamber pressure so ice sublimes directly to vapor without passing through a liquid stage. Secondary drying raises the temperature modestly to remove bound water. The result is a porous, dry solid that usually reconstitutes quickly. Each stage influences pore structure, residual moisture, and stability.
The physics of lyophilization depends on phase boundaries and heat and mass transfer. During primary drying, heat supplied to the product must equal the latent heat of sublimation, while water vapor moves through the drying cake to the condenser. If shelf temperature or pressure is set too high, the ice front can exceed the collapse temperature, causing meltback or pore collapse. If conditions are too cold, drying slows and costs rise. Formulation excipients, vial geometry, and freezing rate alter these limits.
The dissection continues toward the brow and the glabella (the smooth prominence between the eyebrows) until the skin flap is sufficiently mobile to allow its relaxed transposition upon the nose. Under loupe magnification, the distal portion of the forehead flap is de-fatted, down to the subdermal plexus. Yet, the fat-removal should be conservative, especially if the patient is either a tobacco smoker or a diabetic, or both, because such health factors negatively affect blood circulation and tissue perfusion, and thus the timely and correct healing of the surgical scars to the nose. The flap is allowed to perfuse, while the donor site is sutured closed by means of the wide undermining deep to the frontalis muscle. At that time, diluted epinephrine can be injected to the forehead skin, but not to the area(s) near the pedicle of the forehead flap. Moreover, if the distal wound is wider than 25 mm, it usually is not closed by primary intention, with sutures, but is allowed to heal by secondary intention, by re-epithelialisation. The forehead flap is attached to the nasal wound with subcutaneous sutures and skin sutures. If the excess tension of a suture compromises the color of the skin flap, the suture can be loosened, with a skin hook, and observed for 10–15 minutes; if the skin color remains compromised (white), the suture is removed. Upon the complete attachment of the paramedian forehead flap to the nose, the surgical wounds are dressed only with antibiotic ointment. IV.
At the end of the month, it opened a unit in Seoul (owned by 2F Enterprises) and one in Brunei (owned by a subfranchise of A&W Singapore). As of June 1998, A&W only had two restaurants in Mexico. The Cairo branch opened on August 1, 1998, under local franchise American Egyptian Foreign Investments, with Egypt becoming the seventeenth country outside of the United States to open a location. The first co-branded A&W restaurant, tied with Mexican food chain Amigos, opened in Lincoln in 1997. There were plans to increase the amount of co-branded locations across Nebraska by four in the second half of 1998. The goal of the co-branded restaurant was to allure more customers who did not prefer Mexican food. On March 16, 1999, it announced the opening of a restaurant in Dhaka with franchisee Global Supplies Limited due in September that year, with four more planned. The restaurant in Lodi took part in a world record for the largest root beer float of 2562.5 gallons on June 19, 1999, the eve of the eightieth anniversary of the chain. That same year, the chain attempted to enter the Korean market by finding local partners, aiming to grow in the Korean market in the first half of 2000. The plan was ultimately shelved. A&W merged with Long John Silver's to form Yorkshire Global Restaurants based in Lexington, Kentucky in March 1999. Kevin Bazner was promoted president of the chain in July. In 2000, Yorkshire agreed to test multi-branded locations with Tricon Global Restaurants. A grocery location opened in February that year in Idaho.

== Background == Descriptions of rejuvenation and immortality remedies are often found in the writings of alchemists. But all those remedies did not allow even alchemists themselves to live longer than a hundred years. Though the average lifespan of people through the past millennia increased significantly, maximum lifespan almost did not change - even in ancient times there were fairly well and unbiasedly documented cases when some people lived for more than a hundred years (for example, Terentia who lived 103 or 104 years). While among the billions of people of the modern world, there is only one case of life over 120 years (Jeanne Calment, 122 years). The super-long lives of people that are mentioned in ancient books, apparently, are highly exaggerated, since archaeological data show that even the oldest of the ancient people lived no more than modern supercentenarians. In some cases the exaggeration, possibly, is not intentional but occurs due to errors in translation between languages and synchronization of chronological systems. The species limit of human life is estimated by scientists at 125–127 years, and even in the most ideal conditions a person will not live longer due to aging of the body. Some scientists believe that, even if medicine learns how to treat all major diseases, that will increase the average lifespan of people in developed countries by only about 10 years. For example, biogerontologist Leonard Hayflick stated that the natural average lifespan for humans is 92 years.
variable number tandem repeat (VNTR) Any of a class of tandem repeats for which the copy number of the repeated sequence at a particular locus tends to vary between individuals of the same species. VNTRs may occur throughout the genome, both within and outside of coding DNA, and if the copy number is stably inherited may be used in DNA fingerprinting to uniquely identify individuals or to determine their genealogical relatedness to other individuals.

After Banting's death, Best "claimed that the crucial innovation of using alcohol to remove toxic impurities had largely been his own", even though this had actually been Collip's key contribution. In 1972, an official history of the Nobel Committee declared that omitting Best might have been a mistake. In fact, Best was not considered because he was never nominated. Nomination for a Nobel Prize can only be made by certain individuals, including former recipients of the Prize, and his central role along with Banting was not known to those who had the ability to make nominations. Best was subsequently nominated for the 1950 Nobel Prize in physiology based on his work on choline and heparin. At the Centenary Celebration of the Nobel Prize for the Discovery of Insulin held by the Toronto Medical Society on November 27, 2023, Professor Erling Norrby, former chair of the Nobel Assembly at the Karolinska Institute, made a presentation - The Optimal Nobel Prize for Physiology or Medicine and presented the following information from the Nobel archives: “Best was nominated 14 times 1950-1954. The main nominator was Henry Dale who had been supervisor for Best’s Ph.D. work. The discovery proposed to be awarded was Best’s work on the lipotropic effect of choline, but Dale argued that separately that Best should have shared the 1923 prize to Banting. Best was subject to four evaluations by Ulf von Euler who gave support to Dale’s nomination. Although Best was declared worthy of a prize (1951, 1952, 1954) he never received it.”
=== Prenatal testing === Obtaining a prenatal diagnosis for an embryo or fetus in the womb is also possible, using fetal genetic material acquired through chorionic villus sampling. An amniocentesis can be performed if the pregnancy is further along, within 14–18 weeks. This procedure looks at the amniotic fluid surrounding the baby for indicators of the HD mutation. This, too, can be paired with exclusion testing to avoid disclosure of parental genotype. Prenatal testing can be done when parents have been diagnosed with HD, when they have had genetic testing showing the expansion of the HTT gene, or when they have a 50% chance of inheriting the disease. The parents can be counseled on their options, which include termination of pregnancy, and on the difficulties of a child with the identified gene. In addition, in at-risk pregnancies due to an affected male partner, noninvasive prenatal diagnosis can be performed by analyzing cell-free fetal DNA in a blood sample taken from the mother (via venipuncture) between six and 12 weeks of pregnancy. It has no procedure-related risk of miscarriage.
=== Fibrin glue sealant === If blood patches alone do not succeed in closing the dural tears, placement of percutaneous fibrin glue can be used in place of blood patching, raising the effectiveness of forming a clot and arresting CSF leakage.
=== Formulations === Available dosage forms include liquids, syrups, drops, elixirs, effervescent tablets, and powders for mixing with water, capsules, tablets including extended-release formulations, suppositories, compounding powder, and injections.
== Keratin removal == Keratin that is present in the hair, scales and in the epidermis of the skin is hydrolyzed in the presence of alkali (at pH values greater than 11.5). The disulfide bridges found in keratin protein are cleaved but can be reformed. Long periods of liming will result in hair removal. The main removal of keratin is performed during the unhairing operation. In traditional processing, liming and unhairing were indivisible and took place at the same time. During modern liming methods, and in particular the processing of sheepskins, the hair is removed first and then limed in a liming drum. In hair-save technology, the hides are unhaired first and then limed for a further 12–18 hours.

SIMS and FAB are quite similar. SIMS uses an ion beam, usually Ar+ or Cs+. FAB uses a neutral atom beam, usually Ar or Xe. For SIMS and FAB, if the matrix compound is more acidic than the analyte, then predominantly (M + H)+ forms formed, and conversely (M − H)−. Also possibly forming (M + Na)+, (M + K)+, etc, if the matrix is contaminated incidentally (adventitiously) by sodium, potassium, etc. Typical matrix materials include glycerol, thioglycerol, 3-nitrobenzyl alcohol, diethanolamine, triethanolamine, and dithiothreitol-dithioerythritol mixture. They are usually used to analyze polypeptides and oligonucleotides up to 20 kDa. MALDE uses a photon beam, usually the soft-UV 337 nm nitrogen laser. It can also use an infrared (IR) laser for direct analysis of samples contained in gels or thin-layer chromatography (TLC) plates. MALDE can analyze small polymers (~1 kDa), to oligosaccharides, oligonucleotides and polypeptides, antibodies, up to small proteins (~300 kDa). It is highly sensitive, requiring only femtomoles of sample. Desorption/ionization on silicon (DIOS) is similar to MALDE, but without the matrix. The sample is deposited directly on a nanostructured (porous silicon) surface and the sample desorbed directly from the nanostructured surface through the adsorption of laser light energy. DIOS has been used to analyze organic molecules, metabolites, biomolecules and peptides, and, ultimately, to image tissues and cells.
The Ottomans, through the Ma'n dynasty, a great Druze feudal family, and the Shihabs, a mixed Sunni Muslim-Druze family that had converted to Christianity. Ma'n dynasty were a family of Druze chiefs of Arab stock based in the rugged Chouf area of southern Mount Lebanon who were politically prominent in the 15th–17th centuries. Deir al-Qamar was the capital and the residence of the Emirate of Mount Lebanon. The Church of Saidet et Tallé is a Maronite church in Deir el Qamar in Lebanon, it is one of the most important historical and religious sites in Deir el Qamar and dates to the 16th century. The second church was destroyed by the Saracens and rebuilt during Fakhreddine 1st Maan's (1518–1544) reign. In 1673, Sheikh Abu Fares Karam of Ehden (Emir Ahmad Ma'n's secretary) and his brother Sheikh Abu Nader enlarged the church and added a vault. During the reign of Bechir II Chehab (1789–1840) it was again enlarged and renovated. Fakhr-al-Din II (1572–1635) was a Druze prince and a leader of the Mount Lebanon Emirate. For uniting modern Lebanon's constituent parts and communities, especially the Druze and the Maronites, under a single authority for the first time in history, he is generally regarded as the country's founder. Christians prospered and played key roles under his rule, with his main enduring legacy being the symbiotic relationship he set in motion between Maronites and Druze, which proved foundational for the creation of a Lebanese entity. Maronite Abū Nādir al-Khāzin was one of his foremost supporters and served as Fakhr-al-Din's adjutant.
Some types of specialized cell are localized to a particular animal group. Vertebrates for example have specialized, structurally changed cells including muscle cells. The cell membrane of a skeletal muscle cell or of a cardiac muscle cell is termed the sarcolemma. And the cytoplasm is termed the sarcoplasm. Skeletal muscle cells also become multinucleated. Populations of animal groups evolve to become distinct species, where sexual reproduction is isolated. The many species of vertebrates for example have other unique characteristics by way of additional specialized cells. In some species of electric fish for example modified muscle cells or nerve cells have specialized to become electerocytes capable of creating and storing electrical energy for future release, as in stunning prey, or use in electrolocation. These are large flat cells in the electric eel, and electric ray in which thousands are stacked into an electric organ comparable to a voltaic pile. Many animal cells are ciliated and most cells except red blood cells have primary cilia. Primary cilia play important roles in chemosensation and mechanosensation. Each cilium may be "viewed as a sensory cellular antennae that coordinates a large number of cellular signaling pathways, sometimes coupling the signaling to ciliary motility or alternatively to cell division and differentiation." The cilia in other cells are motile organelles, and in the respiratory epithelium play an important role in the movement of mucus.
Austin was contacted by Paul Heyman of Extreme Championship Wrestling (ECW), who had previously managed him in WCW. Heyman hired him to do promos and in-ring interviews as he had not adequately recovered from his injury, paying Austin $500 (equivalent to $1,056 in 2025) a night. Changing his nickname to "Superstar", Austin debuted in ECW at Gangstas Paradise on September 18, 1995. While in ECW, Austin used the platform to develop his future "Stone Cold" persona as well as a series of vignettes running down WCW in general and Bischoff in particular, most memorably in several promos that mocked his then-status as Nitro host by introducing Monday NyQuil, where he was joined by "Bongo" (a set of drums, meant to represent Steve "Mongo" McMichael) in promoting the show "where the big boys play with each other." Several wrestlers have credited ECW as the place where Austin developed his microphone skills. Austin has credited Heyman as the man who taught him how to cut a promo. Whipwreck, who was the ECW World Heavyweight Champion at the time, defeated Austin in an upset to retain the championship at November to Remember on November 18. At December to Dismember on December 9, Whipwreck defended the title against Austin and The Sandman in a three way dance. Austin eliminated Whipwreck from the match by pinning him following a Stun Gun, but was pinned himself after The Sandman punched him using brass knuckles.
Observatories, astronomical – Andean and Mesoamerican astronomers constructed towers to observe the movements of the planets and other astronomical features and events. Although culture groups throughout the world have observed the planets and stars and recorded their movements, the stone structures of the Mesoamerican and Andean culture groups are significant because they show the emphasis these early astronomers placed on making clear and accurate observations. In the U.S., the Anasazi built structures with windows aligned for the observation of celestial events. The most notable example of Maya astronomical observatories is Caracol, in Chichén Itzá. In 1975, archaeoastronomers Anthony F. Aveni and Horst Hartung surveyed the site and suggested that ancient Maya astronomers used the structure to observe the planet Venus. The Maya, as well as other Mesoamerican culture groups, used Venus to set times for ceremonies and as a divination tool.

Most lyophilized products are stored in sealed containers at controlled temperature and humidity. Some require refrigeration, while others are stable at room temperature. Protection from light and moisture is often necessary.
Cake collapse indicates that the porous structure was lost during drying. It can result from excessive product temperature or an unsuitable formulation. Collapsed cakes may have slower reconstitution and are often rejected by visual inspection.
Residual moisture affects the chemical and physical stability of a lyophilized solid. High moisture can promote degradation, aggregation, or cake shrinkage. The acceptable range is set for each product based on stability data.
Freezing only converts liquid to solid. Lyophilization adds vacuum and controlled warming so frozen solvent sublimes, leaving a dry porous solid. The two steps are related but not interchangeable.
