secondary drying raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

After primary drying, secondary drying removes water that remains bound to the material. This stage raises the shelf temperature while maintaining low pressure, which encourages desorption of unfrozen water. Residual moisture can be reduced to a low percentage, improving stability for many products. The process parameters, including freezing rate, shelf temperature, and chamber pressure, influence the final pore structure and reconstitution behavior. Control of these variables helps prevent collapse or meltback during drying.
A formulation often contains excipients that protect the active ingredient during freezing and drying. Bulking agents provide structure, while lyoprotectants stabilize sensitive molecules. The freezing step can produce ice crystals whose size and distribution affect the drying rate, and cycle design includes freezing, annealing, and drying phases. If the product temperature rises above a critical value, the cake may collapse or lose its porous structure. Successful lyophilization therefore depends on the interaction between formulation, equipment, and cycle design.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.

The process usually has three stages: freezing, primary drying, and secondary drying. Freezing sets the ice crystal structure and can determine pore size in the final cake. Primary drying removes free ice by sublimation at low pressure and low shelf temperature. Secondary drying removes bound water by raising the shelf temperature, although some water may remain as part of the solid. Cycle parameters depend on formulation, fill volume, vial type, and equipment performance.
The physical chemistry of freezing influences whether a formulation forms an amorphous glass or a crystalline solid. Amorphous systems can collapse if product temperature rises above the glass transition temperature of the freeze concentrate. Crystalline systems may show eutectic melting, where ice and solute melt together at a fixed temperature. Formulators add bulking agents, lyoprotectants, and buffers to preserve structure and biological activity. The optimum cycle keeps product temperature below critical thresholds during primary drying while allowing efficient sublimation.
Stability programs monitor lyophilized products under defined temperature and humidity conditions over time. Real-time studies at recommended storage conditions are the reference, while accelerated studies provide early signals of degradation pathways. Because a dry cake can still undergo oxidation, hydrolysis, or aggregation, stability depends on residual moisture, excipients, and container headspace. Open questions include how best to predict long-term stability from short accelerated runs and how vial-to-vial variability affects shelf life. Current guidance treats these predictions as product-specific rather than universally generalizable.
Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.
== Summary == Following several years of unsuccessful petitioning through the United Nations and the International Court of Justice for Namibian independence from South Africa, SWAPO formed the PLAN in 1962 with material assistance from the Soviet Union and sympathetic African states such as Tanzania, Ghana, and Algeria. Fighting broke out between PLAN and the South African security forces in August 1966. Between 1975 and 1988, the SADF staged massive conventional raids into Angola and Zambia to eliminate PLAN's forward operating bases. It also deployed specialist counter-insurgency units such as Koevoet and 32 Battalion, trained to carry out external reconnaissance and track guerrilla movements. South African tactics became increasingly aggressive as the conflict progressed. The SADF's incursions produced Angolan casualties and occasionally resulted in severe collateral damage to economic installations regarded as vital to the Angolan economy. Ostensibly to stop these raids, but also to disrupt the growing alliance between the SADF and the National Union for the Total Independence of Angola (UNITA), which the former was arming with captured PLAN equipment, the Soviet Union backed the People's Armed Forces of Liberation of Angola (FAPLA) through a large contingent of military advisers, along with up to four billion dollars' worth of modern defence technology in the 1980s. Beginning in 1984, regular Angolan units under Soviet command were confident enough to confront the SADF. Their positions were also bolstered by thousands of Cuban troops.
=== 1950s === 1950: Isabella Abbott became the first Native Hawaiian woman to receive a PhD in any science; hers was in botany. 1950: Esther Lederberg was the first to isolate lambda bacteriophage, a DNA virus, from Escherichia coli K-12. 1952: Grace Hopper completed what is considered to be the first compiler, a program that allows a computer user to use English-like words instead of numbers. It was known as the A-0 compiler. 1956: The Wu experiment was a nuclear physics experiment conducted in 1956 by the physicist Chien-Shiung Wu, born in China but having become an American citizen in 1954, in collaboration with the Low Temperature Group of the US National Bureau of Standards. That experiment showed that parity could be violated in weak interaction.
According to one report based on a treatment of 60 patients with silibinin, patients who started the drug within 96 hours of ingesting the mushroom and who still had intact kidney function all survived. As of February 2014 supporting research has not yet been published. SLCO1B3 has been identified as the human hepatic uptake transporter for amatoxins; moreover, substrates and inhibitors of that protein—among others rifampicin, penicillin, silibinin, antamanide, paclitaxel, ciclosporin and prednisolone—may be useful for the treatment of human amatoxin poisoning. N-Acetylcysteine has shown promise in combination with other therapies. Animal studies indicate the amatoxins deplete hepatic glutathione; N-acetylcysteine serves as a glutathione precursor and may therefore prevent reduced glutathione levels and subsequent liver damage. None of the antidotes used have undergone prospective, randomized clinical trials, and only anecdotal support is available. Silibinin and N-acetylcysteine appear to be the therapies with the most potential benefit. Repeated doses of activated carbon may be helpful by absorbing any toxins returned to the gastrointestinal tract following enterohepatic circulation. Other methods of enhancing the elimination of the toxins have been trialed; techniques such as hemodialysis, hemoperfusion, plasmapheresis, and peritoneal dialysis have occasionally yielded success, but overall do not appear to improve outcome. In patients developing liver failure, a liver transplant is often the only option to prevent death.
Half-life (symbol t½) is the time required for a quantity (of substance) to reduce to half of its initial value. The term is commonly used in nuclear physics to describe how quickly unstable atoms undergo radioactive decay or how long stable atoms survive. The term is also used more generally to characterize any type of exponential (or, rarely, non-exponential) decay. For example, the medical sciences refer to the biological half-life of drugs and other chemicals in the human body. The converse of half-life is doubling time, an exponential property which increases by a factor of 2 rather than reducing by that factor. The original term, half-life period, dating to Ernest Rutherford's discovery of the principle in 1907, was shortened to half-life in the early 1950s. Rutherford applied the principle of a radioactive element's half-life in studies of age determination of rocks by measuring the decay period of radium to lead-206. Half-life is constant over the lifetime of an exponentially decaying quantity, and it is a characteristic unit for the exponential decay equation. The accompanying table shows the reduction of a quantity as a function of the number of half-lives elapsed.
According to the first option, the spent nuclear fuel is considered complete waste. After an interim storage (wet and dry), all the used fuel is directly disposed of in a deep geological repository. Several geological media and repository designs are being studied according to different natures of fuel and their burnup, radioactive inventory and decay heat generation. The assessment of a geological disposal is based on the multi-barrier approach, which combines multiple effects of engineered and natural barriers in order to delay potential long-lived radionuclide migrations to the biosphere over time. Reprocessing is the alternative option for the spent fuel after a long interim storage. The used fuel is not considered waste anymore but a future energy resource. A large amount of fertile uranium (

He was succeeded by Kazuo Ito as president. In April 2002, Ajinomoto reorganized itself into food, amino acid, and medicine divisions, and owned subsidiaries for frozen foods, fats, and oils. In February 2003, Ajinomoto and Unilever completed a joint venture agreement in six countries and regions of Asia. Because of this, Ajinomoto launched the brand VONO to replace its use of the Knorr brand, and in the process established its own brand identity. In July 2003, Ajinomoto bought the French company Orsan from the UK-based Tate and Lyle Group, renaming Orsan to AJI-NO-MOTO Foods, Europe. In November 2005, AJI-NO-MOTO Pharmaceuticals USA, Inc. was liquidated, and its assets and functions were merged into AJI-NO-MOTO Pharmaceuticals, Europe. In January 2006, Ajinomoto bought the cooking sauce and condiments manufacturer Amoy Food from the French dairy company Groupe Danone. In 2009, the company released "Ajinomoto" to commemorate the 100th anniversary of its foundation.
=== Disinfection === Much research has been done in evaluating the ability of the silver ion at inactivating Escherichia coli, a microorganism commonly used as an indicator for fecal contamination and as a surrogate for pathogens in drinking water treatment. Concentrations of silver nitrate evaluated in inactivation experiments range from 10–200 micrograms per liter as Ag+. Silver's antimicrobial activity saw many applications prior to the discovery of modern antibiotics, when it fell into near disuse. Its association with argyria made consumers wary and led them to turn away from it when given an alternative.
=== Port Adelaide (2015–2019) === At the end of the 2014 season, Ryder left Essendon following the supplements saga. Ryder and his wife Jess were concerned for the health of their unborn child after he was allegedly told of the potential risks of the supplements program when interviewed by ASADA. Of the concern, Ryder stated that "At first we were really scared (about Harlan). What has panned out over a long period of time is that we felt badly let down by the club and lost trust and faith.” He nominated Port Adelaide as his preferred club of destination. Essendon were unwilling to trade Ryder initially, with Ryder and his management suggesting they would take Essendon to the AFL Grievance Tribunal because of the club's breach of its duty of care to players during the scandal. Ryder's manager also suggested that Ryder would consider retirement over returning to Essendon if a trade was blocked by the club. On 16 October, he was traded to the Power. Being Port Adelaide's big name recruit for season 2015, Ryder was unable to take part in the pre-season trial matches and was only cleared from his provisional suspension a few days prior to the round one game against Fremantle due to the ASADA drugs investigation. He played in 18 games for the Power, which included games where he kicked three goals in both showdowns and a four goals against his former club Essendon.
=== Drug demand and decriminalization === According to former Presidents Fernando Henrique Cardoso of Brazil, Ernesto Zedillo of Mexico and César Gaviria of Colombia, the United States-led drug war is pushing Latin America into a downward spiral; Cardoso said in a conference that "the available evidence indicates that the war on drugs is a failed war". The panel of the Latin American Commission on Drugs and Democracy commission, headed by Cardoso, stated that the countries involved in this war should remove the "taboos" and re-examine the anti-drug programs. Latin American governments have followed the advice of the U.S. to combat the drug war, but the policies have had little effect. The commission made some recommendations to United States President Barack Obama to consider new policies, such as decriminalization of marijuana and to treat drug use as a public health problem and not as a security problem. The Council on Hemispheric Affairs states it is time to consider drug decriminalization and legalization. RAND studies released in the mid-1990s found that using drug user treatment to reduce drug consumption in the United States is seven times more cost-effective than law enforcement efforts alone, and it could potentially cut consumption by a third. In 2011, the Obama administration requested approximately $5.6 billion to support demand reduction. This includes a 13% increase for prevention and almost a 4% increase for treatment. The overall 2011 counter-drug request for supply reduction and domestic law enforcement is $15.5 billion, with $521.1 million in new funding.

Its duration of clinical effect is 120–150 minutes, although it is typically administered at 4– to 6-hour intervals. Pethidine has been shown to be less effective than morphine, diamorphine, or hydromorphone at easing severe pain, or pain associated with movement or coughing. Like other opioid drugs, pethidine has the potential to cause physical dependence or addiction. The especially severe side effects unique to pethidine among opioids—serotonin syndrome, seizures, delirium, dysphoria, tremor—are primarily or entirely due to the action of its metabolite, norpethidine.
On December 1, 2024, Biden issued a "full and unconditional" pardon to his son Hunter that covered all federal offenses between that day and 2014, breaking his June 2024 pledge not to do so. In a statement announcing the pardon, Biden said he believed his son was "selectively, and unfairly, prosecuted". On his last full day in office, Biden pardoned five family members, including his brother James, and others targeted for retribution by his successor. They included former chairman of the Joint Chiefs of Staff Mark Milley, former National Institute of Allergy and Infectious Diseases director Anthony Fauci, and members and participants in the House Select Committee on the January 6 Attack. Biden said the pardons were preemptive, to protect the individuals against the incoming Trump administration's politically motivated vengeance, and should not be seen as an acknowledgment of wrongdoing.

Domino's announced on February 7, 2025, that it plans to close more than 200 stores worldwide, 172 of them just in Japan, where Domino's Pizza Japan operates 1,000 stores. The decision comes after CEO and Managing Director Mark van Dyck took the reins of the company. The reason given for the closure was low sales in the selected stores. Of all the markets that include the brand, Australia–New Zealand was the only market to report positive growth. With the closures, Domino's expects to reduce costs by approximately $9.8 million.
Randomized controlled trials show that clonidine monotherapy reduces core ADHD symptoms, including inattention, hyperactivity, impulsivity and disruptive behavior, compared with placebo. Medical reviews on the efficacy of non-stimulant medications for ADHD indicate that clonidine produces moderate effect sizes for core symptom reduction, which are smaller than the large effect sizes reported for psychostimulants. In contrast to the rapid onset seen with psychostimulant medications, clinically significant symptom improvement may be delayed by a few weeks. Reviews of alpha-2 agonists suggest that this drug class may be more effective for managing hyperactivity and impulsivity than for inattentive ADHD symptoms, and that long-term treatment efficacy has been documented more extensively for guanfacine than for clonidine. Unlike psychostimulants, clonidine is regarded as having no abuse potential due in part to a lack of dopaminergic activity along the mesolimbic pathway. Clonidine is also used as an add-on to psychostimulant medications in individuals who have a partial response to psychostimulants, cannot tolerate higher psychostimulant doses, or experience notable evening symptoms. In a randomized controlled trial of ADHD children with an incomplete response to psychostimulants, the addition of clonidine extended-release produced greater reductions in ADHD symptom scores than continuing psychostimulant monotherapy. α2 adrenoreceptor agonists may also improve symptoms in comorbidities of ADHD such as tic disorders, oppositional or aggressive behavior, and insomnia.

Lyophilization removes water by sublimation from a frozen material, while evaporation changes liquid water into vapor. The low-pressure freezing step avoids the liquid phase and can preserve heat-sensitive structures.
Vacuum lowers the pressure below the triple point of water, allowing ice to sublimate directly into vapor. It also helps remove water vapor from the product chamber and shortens primary drying.
Many aqueous solutions and suspensions can be freeze-dried, but some formulations collapse or do not form a stable cake. The process requires careful formulation and cycle development.
Sealed vials or containers should be kept at the temperature specified by stability data, often controlled room temperature or 2–8 °C. Moisture and oxygen barriers are important because both can degrade sensitive materials. Opened containers may need immediate use or protection from ambient humidity.
