After primary drying, secondary drying removes water that remains bound to the material. This stage raises the shelf temperature while maintaining low pressure, which encourages desorption of unfrozen water. Residual moisture can be reduced to a low percentage, improving stability for many products. The process parameters, including freezing rate, shelf temperature, and chamber pressure, influence the final pore structure and reconstitution behavior. Control of these variables helps prevent collapse or meltback during drying.
A formulation often contains excipients that protect the active ingredient during freezing and drying. Bulking agents provide structure, while lyoprotectants stabilize sensitive molecules. The freezing step can produce ice crystals whose size and distribution affect the drying rate, and cycle design includes freezing, annealing, and drying phases. If the product temperature rises above a critical value, the cake may collapse or lose its porous structure. Successful lyophilization therefore depends on the interaction between formulation, equipment, and cycle design.
Analytical methods for lyophilized materials include X-ray diffraction for crystallinity, differential scanning calorimetry for thermal transitions, and scanning electron microscopy for pore morphology. Moisture sorption analysis shows how the cake responds to humidity. These methods help distinguish amorphous from crystalline states and detect phase changes. Open questions remain about how pore structure changes during long-term storage and how best to predict collapse under varied conditions. Comparisons across studies are complicated by differences in formulation, cycle, and storage history.
Quality control for lyophilized products focuses on appearance, moisture level, reconstitution time, and structural integrity. A cake should be uniform, intact, and free of meltback or collapse. Moisture level is measured by Karl Fischer titration or thermogravimetric analysis. Reconstitution time reflects pore structure and formulation. Visual inspection and vial integrity checks detect cracks, stopper defects, or particulate matter. These checks are often performed on samples from each batch. They help confirm that the drying cycle performed as intended.

The low pressure used during drying allows water vapor to move from the ice surface to a cold condenser. Energy supplied as heat drives sublimation but must stay below the collapse temperature of the frozen matrix. If the product becomes too warm, the frozen structure may soften or melt, reducing pore formation and slowing drying. Formulations often include bulking agents, stabilizers, or buffers to support a rigid cake. The final moisture content depends on formulation, freezing rate, and the length of secondary drying.
Freeze-drying is distinct from simple evaporation and from spray drying. Evaporation removes water at temperatures above freezing, while spray drying rapidly dries droplets in a heated gas stream. Lyophilization avoids high temperatures, which can be useful for heat-sensitive materials such as proteins, vaccines, and some foods. The porous cake produced by sublimation dissolves or rehydrates more quickly than a dense dried mass. Not all materials tolerate freezing or the pH shifts that can occur as solutes concentrate during ice formation.
Analytical methods for lyophilized solids must account for the low moisture content and the fragile cake. Karl Fischer titration is widely used for water content, while near-infrared spectroscopy can measure moisture non-destructively in sealed containers. X-ray diffraction and modulated differential scanning calorimetry help identify crystalline or amorphous phases. Residual solvent analysis may be needed if organic solvents were used during formulation. The combination of these methods supports batch release and long-term stability assessment.
Lyophilized products are typically stored as sealed solids in vials or syringes. Moisture ingress is a major concern because many dried cakes are hygroscopic and can lose stability when exposed to humid air. Storage temperature depends on the formulation; some products are kept refrigerated, while others are stable at room temperature. Container closure integrity and headspace moisture are often monitored. Light protection may also be required for some photosensitive materials.
In practice, lyophilization is slower and more energy intensive than simple drying. Cycle times can range from hours to several days depending on load, container, and formulation. Amorphous materials may require excipients that help preserve structure during freezing and drying. The method is widely used for biological materials, pharmaceuticals, and foods where heat drying would cause unacceptable change. Open questions remain about scaling cycles between laboratory and production equipment, and this gap affects technology transfer.
Lyophilization removes water by freezing a material and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intermediate liquid state. Because the material remains frozen during primary drying, the structure often stays porous. This porous matrix can rehydrate quickly when water is added back. The low pressure also allows vapor to leave the solid matrix without boiling.
28 September The 15-year-old girl killed in Croydon, South London the previous day is named locally as Elianne Andam. Police are given an extra 24 hours to question the 17-year-old suspect. The landmark Sycamore Gap Tree, beside Hadrian's Wall in Northumberland, is illegally felled. A 16-year-old boy is subsequently arrested on suspicion of criminal damage. 29 September A school bus carrying 58 people overturns between junction 4 and 5 of the M53 motorway, killing a 15-year-old girl and the driver. A major incident is declared by North West Ambulance Service and Liverpool's Alder Hey Children's Hospital. Dozens of others are treated for injuries, and several are hospitalised. A 17-year-old boy appears in court charged with the murder of Elianne Andam, and is remanded in youth detention to appear before the Old Bailey on 3 October. A second person – a man in his 60s – is arrested in correction with the cutting-down of the world-famous Sycamore Gap Tree. The 16-year-old boy arrested over the incident the previous day is released on bail. In an interview with The Sun, Prime Minister Rishi Sunak reinforces his pro-motorist stance, saying he wants to stop "hare-brained" road calming and safety schemes – including 20 mph zones and low-traffic neighbourhoods – putting an end to what he refers to as a "war on motorists".

==== Other consequences ==== Untreated OSA also leads to a decreased quality of life, difficulties in social functioning, occupational problems, and accidents and a greatly increased rate of vehicle accidents. Those serious outcomes of OSA are mostly related to the excessive daytime sleepiness resulting from the sleep fragmentation and highlight the need to provide the patients with appropriate treatment. Effective treatment majorly improves those adverse consequences, including quality of life. OSA patients also frequently report pain disorders, such as headache or fibromyalgia. OSA patients show an increased pain intensity alongside a decreased pain tolerance.
The ovarian stroma is a unique type of tissue in the ovary that gives structural and metabolic support to the functional parenchyma composed of follicles. The stroma is abundantly supplied with blood vessels, consisting for the most part of spindle-shaped stromal cells. These appear similar to fibroblasts. The stroma also contains ordinary connective tissue such as reticular fibers and collagen. Ovarian stroma differs from typical connective tissue in that it contains a high number of cells. The stromal cells are distributed in such a way that the tissue appears to be whorled. Stromal cells associated with maturing follicles may acquire endocrine function and secrete estrogens. The entire ovarian stroma is highly vascular. On the surface of the organ this tissue is much condensed, and forms a layer (tunica albuginea) composed of short connective-tissue fibers, with fusiform cells between them. The stroma of the ovary contains interstitial cells.

=== Droplet merging === As an existing droplet can be split to form discrete droplets using electrodes (see From an existing droplet), droplets can be merged into one droplet by electrodes as well. Utilizing the same concept applied for creating new droplets through splitting an existing droplet with electrodes, an aqueous droplet resting on an uncharged electrode can move towards a charged electrode where droplets will join and merge into one droplet. However, the merged droplet might not always form a circular shape even after the merging process is over due to surface tension. This problem can be solved by implementing a superhydrophobic surface between the droplets and the electrodes. Oil droplets can be merged in the same way as well, but oil droplets will move towards uncharged electrodes unlike aqueous droplets.
==== Resale exemptions and sales tax (United States) ==== Often, in the United States, wholesalers are not required to charge their buyers sales tax, if the buyer has a resale license in the state the wholesaler is located. Out-of-state buyers are not charged sale tax by wholesalers. Documentation requirements vary by state.
Titanic transformed DiCaprio into a superstar, drawing adoration among teenage girls and young women that became known as "Leo-mania". The success bolstered DiCaprio's standing as a teen idol and romantic lead, an image from which he sought to dissociate himself. Despite the film's success and its positive influence on his career, in 2025 DiCaprio said he regretted the decision to turn down Boogie Nights, saying it was "a profound movie of my generation". In his 2015 Rolling Stone article, Ebiri called the Titanic role DiCaprio's best, writing that he and Winslet "infuse their earnest back-and-forth with so much genuine emotion that it's hard not to get swept up in their doomed love affair". A writer for Vanity Fair in 2008 labeled them "Hollywood's most iconic screen couple" since Humphrey Bogart and Ingrid Bergman. Writing about her first viewing of Titanic in 2017, Vox contributor Alissa Wilkinson described DiCaprio's "boyish charm" and found his performance "natural and unaffected". DiCaprio had a brief featured role in Woody Allen's 1998 satire of fame, Celebrity. Ebiri labeled DiCaprio "the best thing in the film". That year, he also took on the dual roles of villainous King Louis XIV and his secret, sympathetic twin brother Philippe in Randall Wallace's The Man in the Iron Mask, with common elements from the 1939 film and a 1929 film with Douglas Fairbanks. It received mixed reviews, but grossed $180 million against a budget of $35 million.

=== Disease control intervention === Due to the suspicions that food markets could have played a role in the emergence of COVID-19, a group of US lawmakers, NIAID director Anthony Fauci, UNEP biodiversity chief Elizabeth Maruma Mrema, and CBCGDF secretary general Zhou Jinfeng called in April 2020 for the global closure of wildlife markets due to the potential for zoonotic diseases and risk to endangered species. In April 2021, the World Health Organization called for a total ban on the sale of live animals in food markets in order to prevent future pandemics. Planetary health studies have called for disease control intervention measures, in lieu of outlawing live-animal markets, to be implemented in markets. These include proposals for "standardised global monitoring of water, sanitation, and hygiene (WASH) conditions", which the World Health Organization announced in April 2020 that it was developing as requirements for markets to open. Other proposals include less homogeneous policies that are specialized for local social, cultural, and financial factors, as well as new proposed rapid assessment tools for monitoring the hygiene and biosecurity of live animal stalls in markets.
The SeV genome is non-segmented, negative-sense RNA, of about 15.384 n. in length, and contains the noncoding 3′ leader and 5′ trailer regions, which are about 50 nucleotides in length. As in other respiroviruses from family Paramyxoviridae, in SeV they work as cis-acting elements essential for replication. A 3′ leader sequence acts as a transcriptional promoter. Between these non-coding regions are located six genes, which encode the nucleocapsid (NP) protein, phosphoprotein (P), matrix protein (M), fusion protein (F), hemagglutinin-neuraminidase (HN) and large (L) protein in this order from the 3′ terminus. The RNA-dependent RNA polymerase of the SeV consists of the large protein (L) and the phosphoprotein (P). The structural gene sequence of SeV is as follows: 3′-NP-P-M-F-HN-L-5′. Intergenomic regions between these genes are three nucleotides long as in other respiroviruses. Additional proteins, which are frequently called non structural or accessory proteins can be produced from the P gene, using alternative reading frames. The Sendai virus P/C mRNA contains five ribosomal initiation sites between positions 81 and 201 from the 5' end. One of these sites initiates in the P open reading frame, whereas four others initiate a nested set of C proteins (C', C, Y1, Y2). These C proteins are initiated in the + 1 reading frame to that of P at different translation starting sites. Sendai virus uses ribosome shunting to express Y1 and Y2 proteins that initiate at the fourth and fifth start sites on the P/C mRNA (respectively).
Genome-wide analyses of human cancer tissues reveal that a single typical cancer cell may possess roughly 100 mutations in coding regions, 10–20 of which are "driver mutations" that contribute to cancer development. However, chronic inflammation also causes epigenetic changes such as DNA methylations, that are often more common than mutations. Typically, several hundreds to thousands of genes are methylated in a cancer cell (see DNA methylation in cancer). Sites of oxidative damage in chromatin can recruit complexes that contain DNA methyltransferases (DNMTs), a histone deacetylase (SIRT1), and a histone methyltransferase (EZH2), and thus induce DNA methylation. DNA methylation of a CpG island in a promoter region may cause silencing of its downstream gene (see CpG site and regulation of transcription in cancer). DNA repair genes, in particular, are frequently inactivated by methylation in various cancers (see hypermethylation of DNA repair genes in cancer). A 2018 report evaluated the relative importance of mutations and epigenetic alterations in progression to two different types of cancer. This report showed that epigenetic alterations were much more important than mutations in generating gastric cancers (associated with inflammation). However, mutations and epigenetic alterations were of roughly equal importance in generating esophageal squamous cell cancers (associated with tobacco chemicals and acetaldehyde, a product of alcohol metabolism).
=== Full chisel saw chains === The full chisel chain has square-cornered teeth, splitting wood fibers easily in the cut for fast, efficient cutting in clean softwood. Chisel chains have a high kick-back risk due to missing safety chain elements and are more sensitive against dirt. These chains are available for semi-pro (also called pro-sumer) and professional saws.
=== Abortion, sexuality, and family values === In 2016, the Varkey Foundation and Populus conducted an international study examining the attitudes of 20,000 people aged 15 to 21 in twenty countries. They found that just under two thirds of young Americans favored keeping abortion legal, in line with the global average. Gallup polls conducted in 2019 revealed that 62% of people aged 18 to 29—older members of Generation Z and younger Millennials—support giving women access to abortion while 33% opposed. At the national level, Americans are split on the issue of abortion. Many participants in the annual March for Life in Washington, D.C. in the early 2020s are members of Generation Z. However, there is a sex gap in which Gen Z men are significantly less likely to support legal abortion than women and do not think of restrictions on reproductive choices as something important.A 2018 poll conducted by Harris on behalf of the LGBT advocacy group GLAAD found that despite being frequently described as the most tolerant segment of society, people aged 18 to 34—most Millennials and the oldest members of Generation Z—have become less accepting of LGBT individuals compared to previous years.

Lyophilization removes water by sublimation from a frozen material, while evaporation changes liquid water into vapor. The low-pressure freezing step avoids the liquid phase and can preserve heat-sensitive structures.
Vacuum lowers the pressure below the triple point of water, allowing ice to sublimate directly into vapor. It also helps remove water vapor from the product chamber and shortens primary drying.
Many aqueous solutions and suspensions can be freeze-dried, but some formulations collapse or do not form a stable cake. The process requires careful formulation and cycle development.
Karl Fischer titration is a common method, using coulometric or volumetric detection. Thermogravimetric analysis can also measure weight loss on heating. Results depend on sample handling because the dried solid can absorb moisture quickly.
