=== Association methods === Association methods look for characteristic sequences or motifs that can help distinguish between interacting and non-interacting pairs. They look specifically for sequence-signatures that are found together more often than by chance. This uses a log-odds score which is computed as log2(Pij/PiPj), where Pij is the observed frequency of domains i and j occurring in one protein pair; Pi and Pj are the background frequencies of domains i and j in the data. Predicted domain interactions are those with positive log-odds scores and also having several occurrences within the database. The downside with this method is that it looks at each pair of interacting domains separately, and it assumes that they interact independently of each other.
== Biosynthetic pathway == Aflatoxin B1 is derived from both a dedicated fatty acid synthase (FAS) and a polyketide synthase (PKS), together known as norsolorinic acid synthase. The biosynthesis begins with the synthesis of hexanoate by the FAS, which then becomes the starter unit for the iterative type I PKS. The PKS adds seven malonyl-CoA extenders to the hexanoate to form the C20 polyketide compound. The PKS folds the polyketide in a particular way to induce cyclization to form the anthraquinone norsolorinic acid. A reductase then catalyzes the reduction of the ketone on the norsolorinic acid side-chain to yield averantin. Averantin is converted to averufin via a two different enzymes, a hydroxylase and an alcohol dehydrogenase. This will oxygenate and cyclize averantin's side chain to form the ketal in averufin. From this point on the biosynthetic pathway of aflatoxin B1 becomes much more complicated, with several major skeletal changes. Most of the enzymes have not been characterized and there may be several more intermediates that are still unknown. However, what is known is that averufin is oxidized by a P450-oxidase, AvfA, in a Baeyer-Villiger oxidation. This opens the ether rings and upon rearrangement versiconal acetate is formed. Now an esterase, EstA, catalyzes the hydrolysis of the acetyl, forming the primary alcohol in versiconal. The acetal in versicolorin A is formed from the cyclization of the side-chain in versiconal, which is catalyzed by VERB synthase, and then VerB, a desaturase, reduces versicolorin B to form the dihydrobisfuran.

== General overview == Pathologists' assistants work under the indirect or direct supervision of a board certified anatomical pathologist, who ultimately renders a diagnosis based on the PA's detailed gross examination and/or tissue submission for microscopic evaluation. Requirements to become a certified pathologists' assistant include graduation from a National Accrediting Agency for Clinical Laboratory Sciences (NAACLS) accredited education program and successfully passing the American Society for Clinical Pathology (ASCP) certification exam, which is not legally required to perform gross examinations in most states. Some states such as West Virginia, Nevada, and New York require a license for pathologists' assistants. All pathologists' assistants are allied health workers who need to be CLIA 88 compliant to perform these high complexity tasks with indirect/direct supervision of a pathologist. With ongoing changes in health care, a growing population of retiring pathologists, and a decreasing number of pathology residents, well trained PAs are in high demand due to their extensive level of training and contribution to the overall efficiency of the pathology laboratory. === Parkinson's disease === In its oral and ODT forms, selegiline is used to treat symptoms of Parkinson's disease (PD). It is most often used as an adjunct to medications such as levodopa (L-DOPA), although it has been used off-label as a monotherapy. The rationale for adding selegiline to levodopa is to decrease the required dose of levodopa and thus reduce the motor complications of levodopa therapy. Selegiline delays the point when levodopa treatment becomes necessary from about 11 months to about 18 months after diagnosis. There is some evidence that selegiline acts as a neuroprotective and reduces the rate of disease progression, though this is disputed. In addition to parkinsonism, selegiline can improve symptoms of depression in people with Parkinson's disease. There is evidence that selegiline may be more effective than rasagiline in the treatment of Parkinson's disease. ==== Body composition and strength improvements ==== Anabolic steroids influence muscle fiber characteristics, affecting both the size and type of muscle fibers. This alteration significantly contributes to enhanced muscle strength and endurance. Anabolic-androgenic steroids (AAS) cause these changes by directly impacting the muscle tissue's cellular components. Studies have shown that these changes are not merely superficial but represent a profound transformation in the muscle's structural and functional properties. This transformation is a key factor in the steroids' ability to enhance physical performance and endurance. Body weight in men may increase by 2 to 5 kg as a result of short-term (<10 weeks) AAS use, which may be attributed mainly to an increase of lean mass. Animal studies also found that fat mass was reduced, but most studies in humans failed to show significant fat mass decrements. The effects on lean body mass have been shown to be dose-dependent. Both muscle hypertrophy and the formation of new muscle fibers have been observed. The hydration of lean mass remains unaffected by AAS use, although small increments of blood volume cannot be ruled out. === β-Carotene and lung cancer in smokers === Chronic high doses of β-carotene supplementation increases the probability of lung cancer in smokers while its natural vitamer, retinol, increases lung cancer in smokers and nonsmokers. The effect is specific to supplementation dose as no lung damage has been detected in those who are exposed to cigarette smoke and who ingest a physiological dose of β-carotene (6 mg), in contrast to high pharmacological dose (30 mg). Increases in lung cancer have been attributed to the tendency of β-carotene to oxidize, yet based on the pharmacokinetics of β-carotene absorption and transport through the intestine and the lack of specific β-carotene transporters, it is unlikely that β-carotene reaches the lung of smokers in sufficient quantities. Additional research is required to understand the link between the increased risk of cancer and all-cause mortality following β-carotene supplementation. Additionally, supplemental, high-dose β-carotene may increase the risk of prostate cancer, intracerebral hemorrhage, and cardiovascular and total mortality irrespective of smoking status.
