Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Analytical methods for lyophilized materials include X-ray diffraction for crystallinity, differential scanning calorimetry for thermal transitions, and scanning electron microscopy for pore morphology. Moisture sorption analysis shows how the cake responds to humidity. These methods help distinguish amorphous from crystalline states and detect phase changes. Open questions remain about how pore structure changes during long-term storage and how best to predict collapse under varied conditions. Comparisons across studies are complicated by differences in formulation, cycle, and storage history.
Quality control for lyophilized products focuses on appearance, moisture level, reconstitution time, and structural integrity. A cake should be uniform, intact, and free of meltback or collapse. Moisture level is measured by Karl Fischer titration or thermogravimetric analysis. Reconstitution time reflects pore structure and formulation. Visual inspection and vial integrity checks detect cracks, stopper defects, or particulate matter. These checks are often performed on samples from each batch. They help confirm that the drying cycle performed as intended.
Storage stability depends on water content, oxygen exposure, and temperature. Lyophilized solids are hygroscopic and can absorb water if exposed to humid air. Vials are usually sealed under vacuum or inert gas with rubber stoppers and aluminum crimps. Storage temperatures range from room temperature to refrigerated or frozen conditions depending on the material. Stability studies track potency, moisture, and physical form over time. Accelerated conditions can reveal sensitivity but may not predict long-term behavior.
| Property | Value | Notes |
|---|---|---|
| Residual moisture | 0.5-3% w/w | Typical range for many biopharmaceuticals |
| Typical storage temperature | 2-8 °C | Some products require -20 °C or lower |
| Reconstitution time | Seconds to several minutes | Depends on cake porosity and diluent |
| Common moisture method | Karl Fischer titration | Measures water content in the solid |
| Container closure | Stoppered vial with seal | Protects against moisture and oxygen ingress |
Quality control for freeze-dried lots combines visual inspection with instrumental tests. Cake appearance, color, and shrinkage are recorded against a reference, while residual moisture is measured by Karl Fischer titration or loss on drying. Thermal analysis can reveal phase transitions and crystallization events, and X-ray diffraction distinguishes amorphous from crystalline solids. Microbiological tests and container closure integrity checks are also routine for sterile products. Analytical methods must be validated for the matrix, because excipients and low moisture can affect accuracy. Open questions include how best to predict long-term stability from short-term data.
Lyophilized solids are often hygroscopic, so handling occurs in controlled low-humidity areas or glove boxes when the material is exposed. Vials remain sealed with elastomeric stoppers and aluminum crimps until use, because airborne moisture can raise residual water and shorten shelf life. The porous cake is fragile and may crack, shrink, or powder during transport. Personnel typically avoid repeated warming and cooling of sealed units, which can draw moisture through closures. These practices aim to preserve the low water content achieved during drying.
Misconceptions about lyophilization include the idea that dried products are indefinitely stable or that vacuum sealing eliminates all degradation. Chemical reactions can continue in the solid state, and some proteins lose activity even at low moisture. Another misconception is that any freeze-dryer cycle can be scaled by time alone; heat and mass transfer differ with equipment and load. Open questions remain about predicting long-term stability from short accelerated studies, particularly for amorphous formulations. Real-time stability data remain the standard for shelf-life assignment.
After lyophilization, the dried product is often sealed under vacuum or an inert gas to limit moisture and oxygen exposure. Vials, stoppers, and seals must maintain their barrier throughout shelf life. Storage temperature depends on product sensitivity: some cakes tolerate controlled room temperature, while labile biologics require refrigeration. Humidity is a critical variable because dried cakes are hygroscopic and can absorb water when exposed to air. Handling procedures therefore limit open-vial time and use desiccated environments for sampling.
Quality control for lyophilized materials examines appearance, reconstitution time, residual moisture, and mechanical integrity. An acceptable cake is usually uniform and porous, though appearance alone does not prove stability. Karl Fischer titration is a common method for water content, while differential scanning calorimetry can reveal glass transition events. Stability studies track potency, aggregation, and moisture over time under defined temperature and humidity conditions. Specifications are product-specific and may include sterility and endotoxin tests for sterile preparations.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
== Interactions == Concurrent use with other triptans or ergot-containing medications (e.g., ergotamine, dihydroergotamine) within 24 hours can result in additive vasoconstriction. Increased systemic exposure to sumatriptan can occur if used within 2 weeks after a monoamine oxidase inhibitor (MAOI). Cases of serotonin syndrome have been reported with co-administration of triptans and serotonin reuptake inhibitors.

In May 2023, Ajinomoto signed a letter of intent for a strategic alliance with Solar Foods, a company that developed Solein, a microbial protein feeding on CO2 as a nutrient source, and obtained a marketing license in Singapore. In November 2023, Ajinomoto acquired US-based Forge Biologics for $620 million. In September 2024, Ajinomoto entered into a global strategic partnership with Danone to reduce greenhouse gas emissions from Danone’s raw milk supply chain. In the same year, through joint research with Professor Chinatsu Kasamatsu (Project Professor) of the Institute for SDGs Promotion at Ochanomizu University in Japan, Ajinomoto became the first in the world to develop a new concept of 'electric seasoning,' which adjusts the taste of food using transdermal electrical stimulation. In March 2025, Ajinomoto decided to participate as a partner in the 'Demonstration Survey for the Restoration Model of Degraded Agricultural Land in Brazil,' a project under the Japan-Brazil Green Partnership Initiative promoted by the governments of Japan and Brazil. In April 2025, Ajinomoto was selected by CDP, an international environmental not-for-profit charity, for its "Climate Change A-List" for fiscal 2024. This is the fifth consecutive year that Ajinomoto has been selected for the A-list.
=== Argentation chromatography === Ag+ binds alkenes reversibly, and silver nitrate has been used to separate mixtures of alkenes by selective absorption. This phenomenon is the basis of argentation chromatography. In some cases, alkene complexes can be isolated. The alkene can be liberated with ammonia.
Due to the preference for a cis-amide bond with the preceding residue of C2-substituted pseudoprolines, their incorporation results in a kink conformation of the peptide backbone, thus preventing peptide aggregation, self-association, or β-structure formation.

The project intended to stop the flow of firearms from the United States into Mexico and deny cartels weapons considered 'tools of the trade.' In 2011, a gunwalking scandal, later known as "Operation Fast and Furious," occurred when the ATF was accused of permitting and facilitating "straw purchase" firearm sales to traffickers, and allowing the guns to "walk" and be transported to Mexico. Allegedly, the ATF allowed to complete the transactions to expose the supply chain and gather intelligence. It has been established that this operation violated long-established ATF policies and practices and that it is not a recognized investigative technique. Several of the guns sold under the Project Gunrunner were recovered from crime scenes in Arizona, and at crime scenes throughout Mexico, resulting in considerable controversy. One notable incident was the "Black Swan operation" where Joaquín Guzmán Loera was finally captured. The ATF confirmed that one of the weapons the Mexican Navy seized from Guzmán's gunmen was one of the many weapons that were "lost" during the Project Gunrunner. Researchers and Mexican officials have argued that most weapons trafficked into Mexico originate from the United States. The U.S. Department of Homeland Security (DHS) officials have stated that the statistic is misleading: out of approximately 30,000 weapons seized in drug cases in Mexico in 2004–2008, 7,200 appeared to be of U.S. origin, approximately 4,000 were found in ATF manufacturer and importer records, and 87 percent of those—3,480—originated in the United States. The U.S.
Monazite geochronology can also reveal igneous differentiation events such as magma mixing, where the magma chamber is evolved into a different composition. Isomorphous substitution is one of the examples. It is a form of substitution in which one element is replaced by another without changing the crystal structure. In the case of monazite, the rare earth elements are replaced by Ca and Th.
Britain, France, the United States, Canada, and eight other Western European countries signed the North Atlantic Treaty of April 1949, establishing the North Atlantic Treaty Organization (NATO). That August, the first Soviet atomic device was detonated in Semipalatinsk, Kazakh SSR. Following Soviet refusals to participate in a German rebuilding effort set forth by western European countries in 1948, the US, Britain and France spearheaded the establishment of the Federal Republic of Germany from the three Western zones of occupation in April 1949. The Soviet Union proclaimed its zone of occupation in Germany the German Democratic Republic that October. Media in the Eastern Bloc was an organ of the state, completely reliant on and subservient to the communist party. Radio and television organizations were state-owned, while print media was usually owned by political organizations, mostly by the local communist party. Soviet radio broadcasts used Marxist rhetoric to attack capitalism, emphasizing themes of labor exploitation, imperialism and war-mongering. Along with the broadcasts of the BBC and the Voice of America to Central and Eastern Europe, a major propaganda effort began in 1949 was Radio Free Europe/Radio Liberty, dedicated to bringing about the peaceful demise of the communist system in the Eastern Bloc. Radio Free Europe attempted to achieve these goals by serving as a surrogate home radio station, an alternative to the controlled and party-dominated domestic press in the Soviet Bloc.

=== Variants and subgroups === Regulators and clinical trials often use additional subgroups based on how active the disease is, even if there is no firm agreement on their definition. Examples are 'active MS', 'highly active MS' and 'rapidly evolving severe MS'. One definition of highly active MS is having an unchanged or increased number of relapses even after starting disease-modifying treatment. In addition, the term 'relapsing MS' is often used to denote people with relapsing-remitting MS as well as the subset of individuals with secondary progressive MS who still experience some relapses. The terms "benign MS" is sometimes used for people who have had MS for a long time (15 years or so) with few attacks and little disability. As people may still get worse, the term is discouraged. On the other hand, the term aggressive multiple sclerosis (previously known as "malignant multiple sclerosis") is used to describe people with MS having reached a significant level of disability in a short period. Atypical variants of MS have been described; these include tumefactive multiple sclerosis, Balo concentric sclerosis, and Marburg multiple sclerosis.

=== Os cordis === In some animals, the fibrous trigone can undergo increasing mineralization with age, leading to the formation of a significant os cordis (heart bone), or two (os cordis sinistrum and os cordis dextrum, the latter being the larger one). The os cordis is thought to serve mechanical functions. In humans, two paired trigones (left and right) are seen in this essential view of anatomy. As a surgical purchase point, the Trigones risk much in AV propagation. It has been known since Classical times in deer and oxen and was thought to have medicinal properties and mystical properties. It is occasionally observed in many other artiodactyls including goats, sheep, giraffes, camels, water buffalo, and antelopes but also in carnivorans such as otters, dogs, cats, and sea lions and in the horse. It has also been discovered in chimpanzees, the first known great ape to have an os cordis; it has subsequently been reported in a human, though its etiology is unknown. Against the opinion of his time, Galen wrote that the os cordis was also found in elephants. While it is unclear if Galen actually observed the bone, it has since been identified in Asian elephants.
Post-infectious glomerulonephritis can occur after essentially any infection, but classically occurs after infection with the bacteria Streptococcus pyogenes. It typically occurs 1–4 weeks after a pharyngeal infection with this bacterium, and is likely to present with malaise, a slight fever, nausea and a mild nephritic syndrome of moderately increased blood pressure, gross haematuria, and smoky-brown urine. Circulating immune complexes that deposit in the glomeruli may lead to an inflammatory reaction. Diagnosis may be made on clinical findings or through antistreptolysin O antibodies found in the blood. A biopsy is seldom done, and the disease is likely to self-resolve in children in 1–4 weeks, with a poorer prognosis if adults are affected or if the affected children are obese.
The enthalpy of dissolution in hydrochloric acid at standard conditions is −600 kJ/mol, from which the standard enthalpy of formation (ΔfH°) of aqueous Bk3+ ions is obtained as −601 kJ/mol. The standard electrode potential Bk3+/Bk is −2.01 V. The ionization potential of a neutral berkelium atom is 6.23 eV.
== Saturation of the gastric ATPase == Even though consumption of food stimulates acid secretion and acid secretion activates PPIs, PPIs cannot inhibit all pumps. About 70% of pump enzyme is inhibited, as PPIs have a short half-life and not all pump enzymes are activated. It takes about 3 days to reach steady-state inhibition of acid secretion, as a balance is struck between covalent inhibition of active pumps, subsequent stimulation of inactive pumps after the drug has been eliminated from the blood, and de novo synthesis of new pumps.

Residual moisture can influence chemical degradation, cake collapse, and long-term stability. Low moisture levels usually improve stability, but each product has an optimal range.
Most lyophilized products are stored upright at controlled temperatures, often refrigerated or frozen. Protection from moisture and light helps maintain the dried cake.
A diluent is added to the dried cake, which dissolves to form a solution or suspension. Gentle mixing avoids foaming and preserves sensitive molecules.
Karl Fischer titration is a common method, using coulometric or volumetric detection. Thermogravimetric analysis can also measure weight loss on heating. Results depend on sample handling because the dried solid can absorb moisture quickly.
